牛膝根多糖上调单核细胞免疫功能实验 被引量:3

摘要 目的:研究牛膝根多糖(ABPS)在体外对单核细胞活性的调节及诱导单核细胞HLA-DRα表面分子的表达。方法:贴壁法分离培养外周血单核细胞并进行鉴定。ABPS诱导单核细胞后,用电镜技术、中性红实验、四甲基偶氮唑盐(MTT)法分别检测单核细胞超微结构、吞噬能力、增殖的改变;并且用流式细胞术检测单核细胞HLA-DRα表面分子表达水平。结果:ABPS能诱导单核细胞胞浆溶酶体和胞浆量增多、吞噬能力增强,但没有增殖能力;同时上调单核细胞HLA-DRα表面分子的表达。结论:ABPS能增强单核细胞活性,上调单核细胞HLA-DRα表面分子的表达,提示ABPS有增强单核细胞的抗原呈递功能。 OBJECTIVE To study the activated effects of Achyranthes bidentata polysaccharides (ABPS) on human monocytes in vitro, inducing HLA-DRa surface molecules expression of monocytes. METHODS The monocytes were separated by the anchoring technique and identified. After ABPS inducing the monocytes, the ultrastructure, phago-ability, proliferation of monocytes were detected with electron microscope technique, neutral red test, MTT; and HLA-DRa surface molecules expression of the monocytes was detected with flow cytometry. RESULTS ABPS could induce the increasing of lysosomes and plasm number, phago-ability of monocytes with no proliferation; and up-regulate HLA-DRasurface molecules expression of the monocytes. CONCLUSION ABPS can activate monocytes, and up-regulate the HLA-DRa surface molecules expression of the monocytes, providing ABPS increasing immunological function of monocytes.
