病毒性心肌炎MMP-3和TIMP-1的动态变化及其与心肌胶原重构的关系 被引量:5

摘要 目的探讨基质金属蛋白酶-3(MMP-3)和基质金属蛋白酶组织抑制因子-1(TIMP-1)在病毒性心肌炎小鼠发病过程的动态变化及其在心肌胶原重构中的作用。方法50只7周龄雄性Balb/c小鼠腹腔接种0.2 ml的CVB3建立VM模型,应用SP免疫组化法观察VM组心肌炎发病过程中MMP-3,TIMP-1的表达,心脏石蜡切片HE染色计算病理积分,masson三色法胶原特异染色及氯氨T法测定心肌的胶原含量。结果VM小鼠心肌MMP-3和TIMP-1均呈现一过性表达。MMP-3表达于第10天达高峰。TIMP-1于第7天可见表达,第21天达高峰;心肌胶原含量在21,30 d显著增高(P<0.05);MMP-3与心肌病理积分呈正相关(r=0.852,P<0.05),TIMP-1与胶原含量有正相关性(r=0.819,P<0.05)。结论VM小鼠心肌MMPs过度激活并伴有继发的胶原含量增高,且MMPs表达与心肌病理变化密切相关,TIMP-1表达与胶原含量密切相关。MMPs/TIMP参与了心肌炎的病理改变,可能是导致心肌间质改变、胶原增生的因素之一。
