亚砷酸钠处理前后人胚胎皮肤细胞基因表达变化的基因芯片研究 被引量:7

摘要 目的应用基因芯片技术研究亚砷酸钠(NaAsO2)处理前后人胚胎皮肤细胞(HES)基因表达的变化。方法提取NaAsO2处理前后HES细胞的总RNA,纯化为mRNA并反转录成cDNA。分别用Cy3和Cy5两种不同的荧光染料标记成探针,与载有4096个靶基因的表达谱芯片进行杂交,通过扫描、计算机软件分析寻找经NaAsO2作用后差异表达的基因。结果共筛选出包括细胞凋亡、DNA合成修复、原癌基因和抑癌基因等差异表达基因125个,其中65个表达上调,60个表达下调。结论NaAsO2能引起HES细胞内多种基因的表达变化。 Objective To study the differences in gene expressions of HES cells before and after NaAsO2 treatment by the technique of DNA chips. Methods Total RNA of the HES cells before and after NaAsO2 treatment were extracted, purified and subjected to reverse transcription into first-strand eDNA. The eDNA probes were prepared by labeling with Cy3 and Cy5 fluorescence dyes individually and hybridized to the eDNA chips constructed by double-dots of 4096 human genes. Differential expression genes were analyzed by screening and using the computer. Results A total of 125 genes related to apoptosis, DNA synthesis and repair,oncogene and tumor suppressor genes expressed differently after the treatment of NaAsO2, of which 65 genes were up-regulated and 60 genes were down-regulated. Conclusion The expressions of many kinds of genes in HES cells will be altered after NaAsO2 treatment.
