用微量全血彗星试验观察砷中毒病区人群DNA损伤 被引量:9

摘要 目的将微量全血彗星实验方法及微量血样保存方法在地方性砷中毒病区进行实践,以验证该方法的可行性。方法采集病区人群末梢微量血样及相应的水样及尿样,观察人群DNA损伤程度与总砷摄入量及尿砷之间的相关关系以及不同人群DNA损伤程度之间的差别。结果没有发现总砷摄入量及尿砷与DNA损伤程度之间的相关关系,但砷中毒病人的DNA损伤程度明显高于非病人及可疑病人。结论将微量血样保存方法及微量全血彗星实验用于地方性砷中毒病区的人群监测可行。 Objective The comet assay technique and the preservation method of drop whole blood specimen were practised in the area of endemic arsenism to validate their feasibilities. Method The peripheral drop whole blood specimens and the relevant water and urine specimens of the endemic area were collected. The relationships between the degree of DNA damage and the total quantity of ingested arsenic or urine arsenic were studied, and the degrees of DNA damage among different populations in the diseased area were observed. Results The correlation between the degree of DNA damage and the total quantity of ingested arsenic or urine arsenic was not found in this study, but the degree of DNA damage in arsenism group was significantly higher than those in the normal or suspicious case group. Conclusions The preservation method and the comet assay technique of drop whole blood specimen can be applied to the biomonitoring work of the endemic arsenism area.
