摘要
目的研究角膜碱烧伤后修复过程中血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)的表达。方法在建立体内角膜碱烧伤模型的基础上,主要采用免疫组化和RT-PCR等方法以确定VEGF的表达规律。结果VEGF在正常角膜组织和伤后3h几乎无表达。碱烧伤后6h,VEGF开始有明显表达,到伤后12h达到峰值,然后有所下降,碱烧伤后24h和48h表达最低,之后又有一反弹过程,在伤后96h和192h达到最高。表达部位主要集中在基质层。结论VEGF在核酸和蛋白质水平上的表达相似,其表达量随时相点的变化呈波动性趋势。体内角膜组织VEGF的表达是网络调控的结果,与眼新生血管存在着时空对应关系。进一步研究角膜创伤后VEGF对其它抑血管因子的调控作用仍是必需的。
Objective To study the expression of vascular endothelial growth factor (VEGF) in the course of corneal repair after alkali burn in mice. Methods The corneal alkali burn model was established in vivo, then immunocytochemistry (ICC) and RT-PCR were mainly performed to determine VEGF expression in different intervals. Results VEGF could hardly be expressed in both normal tissue and early traumatic tissue of cornea. At 6 h after alkali burn, the VEGF expression levels could be found obviously, then began to increase and reached its maxim at 12 h, then decreased to the minimum between 24 h and 48 h after injury. There existed another increasing process of VEGF expression and reached the second peak at 96 h and 192 h. The VEGF positive reaction mainly gathered in the stroma of cornea. Conclusion The VEGF expression levels in nucleic acid is similar to that in protein, and VEGF expression level showed fluctuating tendency according to the changes of time interval. The result of VEGF expression reflects the network regulation in vivo and in a time- and space-dependent manner with ocular angiogenesis. It is still necessary to further study the role of VEGF in regulating other factors which can inhibit angiogenesis after corneal alkali burn.
