摘要
根据牛Y染色体3个特异基因设计3对引物,分别对牛、牦牛、绵羊、山羊、猪、小鼠基因组进行PCR扩增。3对引物在牛和牦牛基因组中都得到了扩增产物,在绵羊的扩增中只检测到了1个性别决定基因的扩增产物,在其他物种中均没有检测到扩增产物。将其扩增产物进行克隆测序,并采用DNAMAN软件对测定序列进行比对分析,结果表明:3对引物的扩增结果在牦牛和牛的同源性分别达到了99%以上,性别决定基因的扩增产物在绵羊和牛间的同源性达到了94%。
Three pairs of Y chromosome specific primers for different gene in cattle were designed and ap- plied to check genomic DNA of cattle, yak, sheep, goat, pig, mouse respectively. The amplification resuits of all three pairs of primers were obtained from genomic DNA of cattle and yak, only one amplification results of sex determine gene was obtained from sheep genomic DNA, and no amplification result was checked in the other species. After all the amplification products were DNAMAN between cattle and yak or sheep. The identities and yak, the identity of segment of sex determination gene cloned and sequenced, the sequences were blast with of three sequences are more than 99% between cattle is 94% between cattle and sheep.
