幽门螺杆菌对胃上皮细胞株GES-1凋亡及XIAP表达的影响 被引量:4

摘要 目的观察幽门螺杆菌(Hp)慢性感染对胃上皮细胞凋亡及X凋亡抑制蛋白(X IAP)表达的影响。方法通过Hp SS1与人非肿瘤性胃上皮细胞株GES-1共培养,建立GES-1模型细胞,检测模型细胞凋亡率及胃上皮细胞X IAP mRNA的表达。结果Hp感染8周、12周、16周模型细胞凋亡率分别为64.55%、60.58%、45.52%,模型胃上皮细胞的X IAP mRNA实时定量PCR的CT值分别为0.901、0.842、0.796,明显低于对照组细胞凋亡率75.52%(P<0.05)及X IAP mRNA的CT值1.030(P<0.01)。结论Hp慢性感染能抑制胃上皮细胞凋亡,其机制可能与上调X IAP mRNA表达有关。 Objective: To establish a cell model by chronically infecting human gastric epithelial cells with helicobacter pylori (Hp),and to investigate the effect of Chronic Hp infection on apoptosis and expression of X inhibitor of apoptosis protein (XIAP) in gastric epithelial cells. Methods: Human non-tumor gastric epithelial cells GES-1 were cocultured with an Hp strain SS1 ,in order to obtain GES-1 model cells, Apoptosis of the model cells were measured by Annexin V/PI staining and XIAP mRNA expression detected by real-time quantitative PCR, Results: The apoptotic rates of model cells which Hp infected 8 weeks, 12 weeks and 16 weeks (64.55 %, 60. 58%,45.52 % ) were significantly lower than those of control cells (75.52 % ) (P〈 0. 05) ; The CT value of the model cells which Hp infected 8 weeks, 12 weeks and 16 weeks of XIAP mRNA expression detected by real-time quantitative PCR were lower than that of control cells (P〈0.01). Conclusion, Chronic Hp infection induces apoptosis resistance in gastric epithelial cells which is possibly by the mechanism of up-regulating XIAP mRNA expression,
