摘要
采用高效液相色谱法测定枣果中的环核苷酸。色谱条件为:以HYPERSIRC18BDS为固定相,20mmol/LKH2PO4∶CH3OH甲醇(体积比90∶10)为流动相,流速0.6mL/min,温度为室温,检测波长254nm,建立了同时测定枣果中环磷酸腺苷(cAMP)和环磷酸鸟苷(cGMP)的反相高效液相色谱分析方法,并对枣果中cAMP、cGMP进行了全生长期动态分析及对不同品种间cAMP、cGMP的比较分析,明确了各自的变化规律和分布特点。该方法准确、快速、重现性好。
In order to analyse cyclic-nucleotide in jujube, high-performance liquid chromatographic methods was applied. Chromatographic condition: reversed-phase column (4.6mm×200mm) filled with 5μm HYPERSIR C18 BDS as the solid phase; mobile phase was 20 mmol/L KH2PO4: CH3OH (90: 10, V/V); flow rate was 0.6 mL/min and at room temperature. A reversed-phase high-performance liquid chromatographic (HPLC) method for the analysis of cAMP, cGMP in Chinese jujube was developed. Using this analysis methods, changes of cAMP and cGMP during maturity of Chinese jujube were monitored; content of cAMP, cGMP among different cultivars were compared. The method was sensitive, rapid and accurate with good reproducibility.
