TNP-470对人U-251胶质母细胞瘤细胞异种移植瘤生长的影响

摘要 目的探讨TNP-470对人U-251胶质母细胞瘤细胞裸鼠皮下移植瘤生长的影响。方法将人U-251胶质母细胞瘤细胞株(约含1×107个细胞)注射至裸鼠皮下,接种后第7天荷瘤裸鼠随机分为治疗组和对照组,分别于皮下注射等容量的TNP-470和含3%乙醇、5%阿拉伯胶及生理盐水的混悬液,隔日1次,共7次。治疗后第21天测体重及肿瘤大小,以山羊抗小鼠CD105多克隆抗体免疫组化染色计数肿瘤微血管密度(MVD)。结果①治疗后第21天治疗组移植瘤体积[(576.10±114.29)mm3]明显小于对照组[(1512.61±470.25)mm3](P<0.01);抑瘤率为(61.91±6.29)%。②治疗组移植瘤MVD[(5.70±0.85)个/视野]显著低于对照组[(11.32±1.50)个/视野](P<0.05)。③两组裸鼠体重于治疗前后均无显著变化(P>0.05)。结论TNP-470对人U-251胶质母细胞瘤细胞裸鼠皮下移植瘤的生长具有抑制作用而无明显毒副作用。 Objective To investigate the effect of TNP-470 on growth of subcutaneously implanted human glioblastoma U-251 xenografts in nude mice. Methods Twelve nude mice were randomly divided into 2 groups, i.e. treatment group and the control group on the 7th day after the subcutaneous injection of 0.1 ml human glioblastoma U-251 cells (about 1×10^7 cells). In the treatment group and the control group the animals received subcutaneous injection of equal volume of TNP-470 solution (30 mg/kg) and the mixture including 3% ethanol, 5% acacia and 0.9% saline every other day (a total of 7 times) respectively. The tumor size and body weight were determined on the 21st day after the treatment. The tumor microvessel density (MVD) was measured by immunohistochemical technique. Results ①The volume of xenografts in the treatment group [(576.10±114.29)mm^3] was significantly smaller than that in the control group [(1512.61±470.25)mm^3] (P〈0.01). The inhibition rate of tumor growth was 61.91%±6.29% in the treatment group. ②The MVD of xenografts in the treatment group [(5.70±0.85)/HP] was significantly fewer than that in the control group [(11.32±1.50)/HP] (P〈0.05). ③The insignificant change in the body weight of animals were observed after the treatment in both the groups compared with that before the treatment (P〉0.05). Conclusion TNP-470 could significantly inhibit the growth of human glioblastoma U-251 xenografts in the nude mice without evident side effect.
