摘要
目的观察山柰挥发油对人胃癌细胞裸鼠原位移植瘤增殖和转移的影响。方法以人胃癌建系细胞SGC-7901接种裸鼠胃大弯,建立原位移植转移模型,36只荷瘤鼠随机分组用药(对照组、5-FU组、挥发油组、5-FU+挥发油组),12周后收集原位肿瘤淋巴结、肝等组织,计算抑瘤率和肿瘤转移抑制率;免疫组化分析肿瘤微血管密度(MVD)和Ki67蛋白表达情况。结果5-FU组、挥发油组、5-FU+挥发油组抑瘤率分别为34.06%、58.05%和68.22%,各组均有邻近淋巴结转移,肝脏转移率对照组、5-FU组、挥发油组、5-FU+挥发油组分别为88.9%(8/9)、55.6%(5/9)、33.3%(3/9)和11.1%(1/9);MVD、Ki67免疫组化显示挥发油组和联用组MVD、Ki67表达受到明显抑制(P<0.05),且以联用组抑制作用最显著(P<0.05)。结论山柰挥发油有抑制胃癌细胞增殖的作用,并有可能通过抗肿瘤血管生成而起到抑制转移的作用;与化疗药物联合应用有协同增效作用。
Objective:To investigate the effect of volatile oil of Kaempferia galanga L. on the growth and metastasis of human gastric carcinoma in nude mice. nethods:SGC-7901 cells were inoculated below the chorion of the stomach of nude mice to establish the model of metastasis of human gantric cancer. 36BALB-c/nude mice with human gantric cancer were randomized into four groups : control group, 5-FU group, volatile oil group and 5-FU + volatile oil group. After 12 weeks, sample of tumor, lymph nodes and livers were collected and the inhibitary rates of tumor growth and the rates of metastasis were detected. The MVD and expressions of Ki67 were detected by immunohisto-chemical technique. Results : The inhibitary rates of tumor growth in the 5-FU group, volatile oil group and 5-FU + volatile oil group are 34. 06% ,58.05 % and 68. 22% , respectively;The incidences of liver metastases in the four groups are 8/9,5/ 9,3/9 and 1/9. The MVD, Ki67 decreased significantly in volatile oil group and combine treatment group (P 〈 0. 05 ), and the combine treatment group decreased most ( P 〈 0. 05 ). Conclusion : The volatile oil of Kaempferia galanga L can restrain the growth of gastric cancer,and probably by inhibiting tumor angiogenesis to inhibit the metastasis of human gastric cancer. Conbination of the volatile oil of Kaempferia galanga L with cytotoxic agents is more effect.
