黄体酮对脑创伤大鼠学习记忆功能的影响 被引量:4

The effect of progesterone on cognitive function after traumatic brain injury in rats

摘要 目的研究黄体酮对大鼠弥漫性颅脑创伤后皮层、海马区组织水肿及学习记忆功能的影响.方法采用Marmarou的方法建立大鼠弥漫性颅脑创伤模型,将成年Wistar大鼠(300~350 g)32只随机分为脑创伤组、黄体酮治疗组、二甲基亚砜组、假手术组.假手术或脑创伤手术3 d后断头处死,取大鼠脑用干湿重法测皮层和海马组织含水量.采用Y型迷宫于假手术或脑创伤前和处死前检测大鼠学习记忆功能.黄体酮组和二甲基亚砜组大鼠于脑创伤手术后1 h、24 h、48 h,分别给予腹腔注射黄体酮和二甲基亚砜.结果脑创伤后大鼠皮层和海马区组织有明显水肿,而黄体酮治疗后皮层和海马区组织水肿明显减轻(P<0.01);脑创伤组大鼠记忆成绩明显低于假手术组(P<0.05),而黄体酮组大鼠的记忆成绩明显高于脑创伤组(P<0.05).结论黄体酮能够有效地减轻大鼠颅脑创伤后脑组织水肿,明显改善大鼠空间学习记忆功能. Objective To investigate the effect of progesterone(PROG) on water content of cortex and hippocampus after diffuse traumatic brain injury (TBI) and cognitive function in rats. Methods The model of TBI was established according to the method created by Marmarou. Thirty-two Wistar rats (300~350g) were randomly divided into TBI group, PROG group, DMSO group and sham operation group. The rats were executed three days after sham operation or TBI and cortex and hippocampus taken out. Method of dry weight/wet weight was used for assessing water content of cortex and hippocampus. The function of learning and memory was observed by Y-type maze before and after TBI or sham operation in rats. Rats were treated intraperitoneally with PROG and DMSO 1h, 24h and 48h after TBI in PROG and DMSO group respectively, Results There was obvious swelling in cortex and hippocampus in rats of TBI group. Water content of cortex and hippocampus were obviously lighter in PROG group than that in TBI group, The function of learning and memory is obviously lower in TBI group than that in sham operation group, while the function of learning and memory was significantly improved in PROG group compared with TBI group. Conclusion PROG could effectively decrease water content of cortex and hippocampus, and improve space cognitive function.
