摘要
以豫薯4号、豫薯5号、豫薯7号、豫薯12号、豫薯14号为材料,以M S为基本培养基对甘薯茎尖分生组织进行不同激素种类和浓度试验,结果表明,最佳诱导分化培养基为:M S+6-BA l.0m g/L+NAA 0.1m g/L+GA30.05m g/L;继代培养与生根培养用.M S+NAA 0.2m g/L为好。在28℃、光照14h/d条件下培养效果好。试管苗株系经指示植物、NCM-EL ISA法检测,获得了5个品种的脱毒苗。
Stem tip meristems of 5 good varieties of sweet potato (Ipomoea potato Lam. ),including Yushu 4 ,Yushu 5 ,Yushu 7,Yushu 12,Yushu 14,were used as explants and cultured on MS medium supplemented with different combinations of hormones at different concentrations. The optimum medium for differentiation induction was MS+6-BA 1.0mg/L +NAA0.1mg/L +GA3 0. 05mg/L. The optimum medium for rapid propagation and rooting was MS+NAA0. 2mg/L ; Results of meristem culture was satisfactory in the conditions of 28℃ and illumination 14 h/d. Five virus-free varieties were obtained after tube derived lines were detected by means of the test of indicator plants and NCM-ELISA.
