摘要
建立了山药中尿囊素的高效液相色谱分析方法。样品用甲醇-水(90:10,V/V)提取,在氨基柱上进行高效液相色谱分析,紫外检测波长224nm,流动相为甲醇-水(80:20,V/V),相对标准偏差为2.0%(n=5),尿囊素的回收率为98.8%(n=4)。
The determination of allantoin was aimed at the quality control for the extract of dioscorea opposta thunb. In this paper,a new HPLC method for this purpose has been developed. The effects of extracting solvent. extracting time, pretreatment of sample, packing materials and composition of mobile phase on the separation of allantoin and the analytical results were investigated. The samples were ultrasonically extracted with methanol water (90: 10 , V/V)for 20min. After standing for 30 min the suspension was centrifuged (5min at 2000 rpm) and the supernatant was injected into HPLC system. The conditions of HPLC were: column: Shim-pack CLCNH2 (150×6.0mm i. d. ) at ambient; mobile phase: methanol-water (80: 20, V/V)and detector: UV at 224nm. The calibration curve was linear in the range of 0.016-0.064μg of allantoin with correlation coefficient of 0.9990. Relative standard deviation was 2.0%(n=5)and recovery of allantoin was 98. 8% (n=4 ).
