摘要
目的:观察藻鳖软肝汤对实验性肝纤维化大鼠肝脏基质金属蛋白酶(MMP-1)、基质金属蛋白酶抑制物(TIMP-1)、血小板源性生长因子(PDGF-β)及肝组织病理学变化的影响,探讨其抗纤维化作用机制。方法:60%CCl4经大鼠皮下注射制备纤维化模型,按组分别给生理盐水,小、中、大剂量藻鳖软肝汤治疗。用光镜观察肝病理变化,免疫组化染色法检测PDGF-β、MMP-1、TIMP-1。结果:藻鳖软肝汤能显著改善肝纤维化大鼠肝脏病理变化(P<0.05),抑制肝细胞变性坏死及纤维结缔组织增生;PDGF-β、TIMP-1及TIMP-1/MMP-1均显著下降(P<0.05),MMP-1无明显变化。结论:藻鳖软肝汤抑制PDGF-β和TIMP-1的合成,调节TIMP-1/MMP-1比值,阻止胶原的生成,促进胶原的分解,抑制肝细胞变性坏死及结缔组织增生,达到抗纤维化的目的。
Objective: To study the effects of Chinese traditional medicine Zaobiemangan decoction on fibrosis of liver in rats. Methods: Creating models of fibrosis of liver in rats by 60% CCl4, then, comparing changs PDGF-β, MMP-1, TIMP-1, TIMP-1/MMP-1 and pathogenic hisology in liver between model-group and study-group. Results: Zalbiemangan decoction can reduce the activity of PDGF-β, TIMP-1, TIMP-1/MMP-1. It also restrains necrosis of liver cell and fibrosis of liver. Conclusion: Chinese traditional medicine Zaobieruangan decoction can prevent and treat fibrosis of liver in rats.
