摘要
[目的]建立有效消除光谱吸收峰不对称给定量分析造成的影响,并校正基于干扰组分的吸收光谱具有线性吸收产生的基线倾斜的方法.[方法]本实验采用三波长紫外分光光度法.[结果]线性回归方程为:△A=-0.02048+122.64865*C,相关系数r=0.99982.说明总黄酮的浓度在0.002~0.0042mg/mL范围内,△A与浓度C之间呈良好的线性关系,可按标准曲线进行定量分析.[结论]本方法的平均回收率为98.76%~101.97%,变异系数小于1%.而且,本法操作简便、快速、准确,具有良好的分析应用前景.
A method was developed to determine total flavonoids in Glycyrrhiza to eliminate the interfering by the asymmetric absorbance error and to correct the scattering effect by the linear absorbance. Three - wavelength Spectrophotometry was used. ΔA = - 0.02048 + 122. 64865 * C, r= 0.99982. It shows that the concentration of total flavonoids is 0.002 - 0.0042mg/mL. ΔA and concentration C show good linear connections which can be quantitative analyzed by standard curves. The average recoveries are 98.76- 101.97%, and the differential is ≤ 1%. The method is simple, rapid and accurate with good prosperity.
