银杏叶提取物对实验性大鼠肝纤维化的预防作用 被引量:14

摘要 目的研究银杏叶提取物(EGB)对肝纤维化大鼠TGFβ1影响及抗纤维化作用。方法30只雄性SD大鼠被随机分为三组:正常对照组(10)、模型组(10)、及EGB干预组(10),模型组及EGB干预组给予50%CCL4腹腔注射,1ml/kg,1周两次,共8周,干预组每天同时给予EGB灌胃,0.4g/kg。实验结束后,处死动物分离血清行肝功能生化指标检测,取肝脏液氮冻存及甲醛固定,分别行HE及Masson染色,免疫组化检测TGFβ1和αSMA,RTPCR检测TGFβ1mRNA的表达。结果生化指标检测EGB干预组优于模型组(P<0.01),光镜下组织学检查纤维化分级、图像分析系统测量胶原面积EGB干预组低于模型组(P<0.05),免疫组化检测EGB干预组TGFβ1和αSMA的表达低于模型组(P<0.01),RTPCR检测EGB干预组TGFβ1mRNA的表达低于模型组(P<0.01)。结论EGB对CCL4诱导的大鼠肝纤维化有良好的防治作用。 Objective To investigate the effects of EGB on hepatic fibros h and expression of TGF-β1 in rats with cirrhosis. Methods Thirty adult male SD rats randomized into 3 groups: control group (n = 10),model group (n = 10) and treatment group(n = 10). Except the rats of control group, others were inflicted with liver cirrhosis by administration of CCIA (50%, 1ml/kg) for 8 weeks. Those rats in the treatment group were intragastrically administered with EGB(0.4g/kg) for 8 weeks. At the end of the 8th week, all rats were sacrificed. Blood samples were collected for determination of ALT, AST and ALB. The area of collagenwas examined by image analyzing system. The expression of TGF-β1 and α-SMA was deter mined by immunohistochemistry. The expression of TGF-β1 mRNA was deter mined by RT - PCR. Results The level of ALT, AST and ALB in EGB-treated group were lawerthan that of model group ( P 〈 0.01 ). The grade of fibrosis and the area of collagen in EGB-treated group were lower than that of the model group ( P 〈 0.05). The treatment with EGB significantly reduced mRNA and protein levels of TGF-β1, and also reduced protein levels of α-SMA. Conclusion EGB can significantly attenuate the degree of hepatic fibrosis.
