摘要
目的:探讨细胞周期素E(Cyclin E)、p27kip1蛋白在大肠癌中的表达及意义.方法:应用免疫组化S-P法检测85例大肠癌、20例大肠腺瘤标本中Cyclin E、p27kip1的表达,并分析与大肠癌临床病理特征的关系.结果:随着大肠粘膜病变由良性向恶性的转变,Cyclin E表达呈逐渐升高趋势,并与大肠癌Dukes分期、淋巴结转移显著正相关,与p27kip1表达负相关;p27kip1表达呈逐渐降低趋势,与大肠癌Dukes分期、淋巴结转移、肠壁浸润及Cyclin E表达呈负相关.结论:Cyclin E和p27kip1是大肠癌恶性程度的正性和负性相关因子,检测二者表达情况有助于判断大肠癌的恶性趋势及生物学行为,并有助于估计患者的预后,为临床治疗提供依据.
Objective: To investigate the expression of Cyclin E, p27^kip1 protein and the clinicopatho-logical significance in colorectal carcinomas. Methods: The expression of Cyclin E,p27^kip1 protein was detec-ted with immunohistochemical stain technique (S-P method) in the paraffin embeded tissue samples (85 cases of colorectal carcinoma, 20 cases of adenorna and 20 cases of normal colorectal mucosa). The data related to colorectal carcinoma clinicopathological characteristics was analyzed. Results: With the transformation of colorectal mucosa from benign to malignant status, the expression of Cyclin E was upregulated gradually, and significantly correlated with Dukes stage, lymph nodal metastasis, but has inverse correlation with the expression of p27^kip1. Meanwhile, p27^kip1 expression appeared to be downregulated, and had an inverse correlation with Dukes stage, lymph nodal metastasis, deep infiltration and the expression of Cyclin E. Conclusion: Cyclin E and p27^kip1 are the positive and negative correlative factors implying the malignant degree of colorectal carcinoma respectively; detecting the expression of Cyclin E and p27^kip1 is helpful for evaluating the malignant degree and biological behavior of colorectal carcinoma, and for evaluating the prognosis of patients. It also prorides the evidences for clinic treatment.
