摘要
目的观察大鼠脑细胞内钙浓度([Ca2+]i)的变化及βAP1-42(β-amyloidproteinfragment1-42)对[Ca2+]i的影响。方法以Fura/AM为荧光指示剂,采用荧光分光光度计测定急性分离的大鼠脑细胞[Ca2+]i。结果静息状态大鼠脑[Ca2+]为304±35mmol.L-1,高钾去极化则使细胞内钙明显增加,βAP对高钾去极化下[Ca2+]i的增加有明显增强作用(P<0.01)结论βAP在某些情况下能使动物脑细胞内钙更明显超载。
Objective To observe the change of intracellular calcium ([Ca^2+]i) in rat brain cells and the effect of β-amyloid protein on [Ca^2+]i Method Fluorescence spectrophotometer with fura-2 as a calcium fluorant was used to measure [Ca^2+]i of acutely dissociated brain cells from rat. Result In the presence of extracellular Ca^2+ 1mmol·L^-1, resting [Ca^2+]i was 304±35mmol· L^-1 ,the response of rise in [Ca^2+]i to KCl in brain cells was significant. β-amyloid protein fragment 1-42 (βAP1-42) significantly enhanced the increase of [Ca^2+]i induced by different concentration of KCl. Conclusion βAP significantly increases [Ca^2+]i of animal brain in some settings.
