摘要
目的了解Bel-7402与QGY-7701人肝癌细胞株生长激素受体(GHR)的表达情况,以获得肝癌GHR在细胞水平上的定量表达。方法分别采用放射性配体分析与RT鄄PCR法检测GHR在Bel鄄7402与QGY-7701肝癌细胞株的表达。结果7402肝癌细胞可表达GHRmRNA,在蛋白表达水平放射性配体法发现GHR在此肝癌细胞位点数量(Site,103/cell)为7.348±0.891,平衡解离常数为0.63±0.04583nmol/L;未发现7701肝癌细胞株表达GHR。结论7402肝癌细胞株表达GHR,可为将来研究rhGH与原发性肝癌的增殖关系奠定一些实验基础。
Objective To Investigate the expression of growth hormone receptor(GHR) on Bel-7402 and QGY-7701 human hepatocellular carcinoma cell strains, thus to get quantitative expression of GHR of hepatocellular carcinoma on the cell level.Methods The expression of GHR on Bel-7402 and QGY-7701 human hepatocellular carcinoma cell strains were detected by radioligand receptor assays and RT-PCR.Results GHRmRNA was expressed on 7402 human hepatocellular carcinoma cell strain, at protein level radioligand receptor assays showed that the site of GHR on this hepatocellular carcinoma cell strain was 7.348±0.891,while the balanced dissociation constant(Ka) was 0.63±0.04583 nmol/L.No GHR expression was found on QGY-7701 human hepatoeellular carcinoma cell strain.Conclusion GHR can be expressed on 7402 human hepatoeellular carcinoma cell strain. This finding might be significant in the research of the multiplied relationship between rhGH and hepatocellular carcinoma.
