摘要 目的探讨白细胞介素4(IL-4)及白细胞介素4受体(IL-4R)基因多态性与郑州地区儿童哮喘易感性及与血浆总IgE的关系。方法采用聚合酶链反应-限制性酶切多态性方法((PCR-RFLP)检测IL-4基因启动子区C-589T位和IL-4Rα亚单位576QR两位点基因多态性,并观察对血浆总IgE的影响。结果白细胞介素4基因C-589T基因多态性与儿童哮喘无相关性;IL-4受体α亚单位576QR基因型和576R等位基因频率在儿童哮喘与对照组相比差别有显著性(P<0.01,P<0.01),且576R与高IgE相关。结论本研究表明IL-4Rα亚单位576R与中国郑州儿童哮喘有关,且与高IgE相关。 Objective To evaluate the possible relationship between asthma and polymorphisms of IL-4 receptor (576QR) and IL-4 (C -589T)and serum IgE in children of zhengzhou area. Methods Two polymorphism sites of IL - 4 and IL - 4R were studied by polymerase chain reaction- restriction fragment length polymorphism( PCR - RFLP) . Results The IL - 4 (C - 589T) was not associated with children asthma, however, the IL - 4Rα. The results showed that chain 576QR genotype and 576 R allele were significantly increased in the subjects with asthma in children compared with subjects ( P 〈 0.01, P 〈 0.01 ) . The IL - 4Rα chain 576R genotype was associated with higher plasma IgE. Conclusion IL - 4Rα polymorphism (576R) is associated with the development of asthma and is also related with higher plasma IgE level.
