PTEN、VEGF蛋白表达与乳腺癌病理学特征以及微血管密度的关系 被引量:3

摘要 目的探讨乳腺癌组织中PTEN、VEGF蛋白表达与临床病理学特征的关系及其与微血管密度(MVD)的相关性。方法采用SP法检测42例乳腺癌组织及15例正常乳腺组织中PTEN、VEGF和CD34蛋白的表达。结果PTEN蛋白在正常乳腺组织高表达率为80%(12/15),在乳腺癌组织中的高表达率为35.7%(15/42),两者比较有显著性差异(P<0.01);乳腺癌组织中PTEN蛋白表达与患者年龄、肿瘤大小、临床分期无关(P>0.05),而与淋巴结转移、组织学分级有关(P<0.05);VEGF蛋白在正常乳腺组织高表达率为20%(3/15),在乳腺癌中高表达率为88.1%(31/42),两者比较有显著性差异(P<0.01);乳腺癌组织中VEGF蛋白表达与患者年龄、肿瘤大小、临床分期和组织学分级无关(P>0.05),而与腋淋巴结转移有关(P<0.05);MVD值在PTEN高表达、VEGF低表达的组织中明显低于PTEN低表达、VEGF高表达的组织,两者比较差异显著(P<0.05)。结论联合检测PTEN、VEGF和MVD的表达可作为乳腺癌生物学行为和预后判断的重要指标。 Objective To investigate the relationship among the PTEN expression,VEGF expression, microvessel density (MVD)and the clinicopathological feattares of breast cancer. Methods The expression levels of PTEN,VEGF and CD34 protein were assessed in 42 cases of breast cancer tissues and 12 cases of normal breast tissues with SP immunohistochemical method. Results The over-expression rate of PTEN protein were 80 % (12/15) in normal breast tissues and 35. 7 % (15/42)in breast cancer cases respectively with a significant difference between two groups(P 〈0.01). Expression of PTEN was not related to age,tumour size and clinical stage(P〈0. 05), but closely correlated to lymph node metastasis and histologic grade(P〈0.05). The over-expression rate of VEGF protein was 88. 10%(31/42)in the breast cancer tissues,and 20^(3/15)in normal breast tissues which had significant difference between two groups(P〈0.01). Expression of VEGF was not related to age, tumor size, clinical stage and histologic grade(P〈0.05), but closely correlated with maxillary lymph node metastasis(P〈0.05). The MVD of the breast cancer tissues with low expression of PTEN or high expression of VEGF was obviously higher than that of the tissues with high expression of PTEN or low expression of VEGF(P〈0. 05). Concusion The combination detection of PTEN,VEGF and MVD could serve as an important index to estimate the biological behavior and prognosis of breast cancer.
