实时荧光定量PCR法检测BARF1基因在鼻咽癌中的表达 被引量:3

摘要 目的通过检测EB病毒编码的BARF1基因在鼻咽癌组织中的表达水平,探讨BARF1基因与鼻咽癌之间的关系。方法应用实时荧光定量PCR法对BARF1基因在鼻咽癌、头颈部其他恶性肿瘤及慢性鼻咽炎病人组织中的表达进行检测。结果BARF1基因在鼻咽癌、头颈部其他恶性肿瘤及慢性鼻咽炎组织中的阳性表达率分别为100%、94.4%和80%,三者组间进行比较差异无统计学意义(P>0.05);鼻咽癌组织中BARF1基因的相对表达强度中位数为11.87,头颈部其他恶性肿瘤为0.07,慢性鼻咽炎组织为0.02。BARF1基因在鼻咽癌组织与头颈部其他恶性肿瘤及慢性鼻咽炎组织中的表达强度进行比较,均有统计学意义(P<0.05)。结论BARF1基因在各种组织中存在着普遍的表达,而在鼻咽癌组织中存在着较高的表达强度,提示BARF1基因在鼻咽癌细胞癌变过程中可能发挥重要作用。 Objective To detect the expression level of BARF1 gene encoded by EBV in NPC tissue, and to investigate the relationship between 13ARF1 gene and NFC. Methods BARF1 expression levels in NFC, non-NPC head and neck tumors, and chronic nasopharyngitis were detected by Real-time PCR analysis. Results All NPC samples, 94.4% non-NPC head and neck tumors, and 80% chronic nasopharyngitis had detectable BARF1 expression. There was no significant difference among them ( P 〉0.05). The median of BARF1 expression level was 11.87 in NPC, 0.07 in non-NPC head and neck tumors, and 0.02 in chronic nasopharyngitis. The expression level of BARF1 in NPC tissue was significantly higher than those either in non-NPC tumors or in nasopharygitis ( P 〈0.05). Conclusion BARF1 gene may play an important role in the genesis of NPC.
