缺氧诱导因子-1α、VEGF在进展期胃癌中的表达及意义 被引量:11

Expression and pathobiological implication of hypoxia-inducible factor-1α and vascular endothelial growth factor in human advanced gastric carcinoma

摘要 目的揭示进展期胃癌中缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)的表达规律,分析其与胃癌临床病理特征的关系,以探讨HIF-1α蛋白在胃癌血管形成中的作用。方法应用免疫组化方法检测50例胃癌组织HIF-1α、VEGF蛋白及微血管密度(MVD)的表达。结果胃癌组织HIF-1α蛋白与VEGF阳性表达率分别为68%(34/50)和62%(31/50),MVD平均计数为23.14±11.58;HIF-1α表达与淋巴结转移有关(P<0.05);VEGF、MVD表达与肿瘤浸润深度(P<0.05)、淋巴结转移(P<0.05)和PTNM临床分期(P<0.05)密切相关;HIF-1α与VEGF表达密切相关(χ2=4.96,P<0.05);HIF-1α或VEGF表达阳性的MVD值显著高于阴性组(P<0.05)。结论HIF-1α、VEGF是反映胃癌浸润转移等生物学行为的重要指标,HIF-1α诱导VEGF表达从而促进肿瘤血管生成可能参与胃癌的进展。 Objective To study the expression of hypoxia - inducible factor- 1α (HIF- 1α) and vascular endothelial growth factor (VEGF) in advanced gastric carcinoma and to analyze their relationship with clinicopathology Characters in order to determine their effect on angiogenesis. Methods lmmunohistochemistry was used to detect the protein expression of HIF- 1α, VEGF and microvessel density (MVD) in gastric carci- noma. Results The positive rates of HIF- 1α and VEGF protein expression in patients with advanced gastric carcinoma were 68% (34/50) and 62.0% (31/50) respectively, MVD was 23.14 ± 11.58. The expression of HIF- 1α was positively related to lymph node metastasis ( P 〈 0.05) ; VEGF and MVD was positively correlated with tumor invasion ( P 〈0.05), lymph node metastasis ( P 〈 0.05) and pTNM stage ( P 〈 0.05). Ex- pression of HIF- 1α was significantly related with the expression of VEGF (x^2 = 4.74, P 〈 0.05) ; MVD was significantly higher in GUF- 1α and/or VEGF positive tumors than those in negative groups ( P 〈 0.05). Conclusion HIF- 1α or VEGF expression was one of the predictors for the biological behavior of advanced gastric carcinoma. HIF- 1α may play an important role in angiogenesis and tumor progression by regulating the expression of VEGF in human advanced gastric carcinoma.
