应用正向间接血凝试验检测山羊痘抗体 被引量:2

摘要 采用硫酸铵盐析、Sephadex G-200柱层析的方法纯化山羊痘病毒,以病毒抗原致敏绵羊红细胞建立了山羊痘正向间接血凝试验,并进行了最佳试验条件的摸索。结果表明,试验所制备的诊断液能与山羊痘参考阳性血清发生特异性凝集,其凝集现象能被山羊痘阳性抗原特异性抑制,与其它常见的动物病阳性血清无交叉反应。用本方法及琼脂扩散试验对山羊痘血清进行检测,IHA阳性检出率为89.1%(41/46);而AGP阳性检出率为34.8%(16/46)。表明正向间接血凝试验比琼脂扩散试验敏感性高。本次试验制备的正向间接血凝试验诊断液可用于兽医临床上进行快速诊断。 The goat pox virus(GTPV) has been purified through ammonium sulfate sedimentation and Sephadex G -200 chromatography. The sheep erythrocyte was sensitized with purified virus and the indirect hemagglutination test (IHA) was developed. At the same time, the optimal conditions for the experiment have been surveyed. This diagnostic reagent could specifically reacted with reference positive serum of goat pox. Furthermore the bemagglutination reaction could be specifically inhibited by GTPV antigen and no cross reaction was observed with other common sera of animal virus diseases. The serum samples of affected goat were detected by IHA and agar gel precipitation (AGP). The result indicated that IHA was more sensitive than AGP and the reagent could be used for diagnosing goat pox quickly.
