红花黄素预处理对缺血再灌注大鼠心肌细胞凋亡的影响 被引量:16

摘要 目的研究红花黄素预处理对心肌缺血-再灌注大鼠心肌细胞凋亡的影响。方法采用在体左冠状动脉前降支结扎法制备心肌缺血-再灌注模型。24只SD大鼠分为3组:假手术组,缺血-再灌注组,红花黄素预处理组。缺血30 min,再灌注60 min。采用缺口末端标记法以及免疫组化法,观测心肌细胞凋亡指数(AI)、Bcl-2、Bax基因蛋白表达。结果与假手术组比较,缺血-再灌注组AI增加,Bax阳性表达增强,Bcl-2阳性表达减弱(P<0.01);与缺血再灌注组比较,红花黄素预处理组AI、Bax阳性表达下降(P<0.01),Bcl-2阳性表达上调(P<0.05)。结论红花黄素预处理通过抑制心肌细胞凋亡,下调Bax基因蛋白表达,上调Bcl-2基因蛋白表达,起到心肌缺血再灌注损伤的保护作用。 Objective To study the effects of safflor yellow (SY) on myocardial apoptosis induced by myocardial ischemia reperfusion in rats. Methods Myocardial ischemia reperfusion injury model was induced by the ligation of left descend coronary artery (LAD) for 30 min, followed reperfusion for 60min in vitro. 24 SD rats were divided into three groups: sham group, I/R group and SY group. Apoptsis index (AI) of myocardium was detected by TUNEL method and protein expression of Bcl-2 and Bax genes was investigated by S-P immuno-hisochemial stain. Results In I/R group, AI was significantly increased, where a protein expression of Bcl-2 gene was decreased and protein expression of Bax gene was increased. SY could decrease AI in I/R group (P〈0.01) ,up-regulating protein expression of Bcl-2 gene(P〈0.05), and down regulate protein expression of Bax gene(P〈0.01 ). Conclusion Injection of SY can protect myocardium damage induced by I/R injury. The mechanism of cardio protection may be associated with inhibiting apoptosis of myocardium, up-regulating protein expression of Bcl-2 gene and down regulating protein expression of Bax gene.
