冬虫夏草对糖尿病肾病鼠肾组织转化生长因子β/C-myc表达的影响 被引量:28

摘要 目的:①研究糖尿病肾病(DN)模型鼠肾组织转化生长因子β(TGF-β)与C-myc的表达状况及C-myc基因在DN发病中的作用和意义;②研究冬虫夏草对TGF-β与C-myc基因表达的影响。方法:45只Wistar大鼠随机分为正常对照组N组,DN模型组D组,冬虫夏草干预组C组[冬虫夏草5·0g/(kg·d),灌胃]。第2、4、6周各组处死5只并收集标本,检测24h尿蛋白定量、血肌酐、血胆固醇、血三酰甘油、血TGF-β、血Col-Ⅳ及肾脏病理改变,应用免疫组化法检测肾组织TGF-β与C-myc基因的表达,并分析TGF-β与C-myc之间的相关关系。结果:C组24h尿蛋白定量低于D组(P<0·01),血肌酐、血胆固醇、血TGF-β、血Col-Ⅳ均低于D组(P<0·05),肾脏病变减轻,免疫组化显示D组TGF-β与C-myc基因的表达高于N组(P<0·05),C组TGF-β与C-myc基因的表达低于D组(P<0·05)。结论:冬虫夏草抑制肾组织TGF-β与C-myc基因表达,对早期DN可能具有保护作用。 Objective:To investigate the expression TGF-β and C-myc gene on renal tissues in STZ rats, and the relationship of C-myc gene ( one of protooncogene) in the pathogenesis of diabetic nephropathy, the relationship between TGF-β and C-myc gene, and the influence on TGF-β and C-myc gene of cordyceps sinensis(CS). Methodology:Fortyfive Wistar rats were divided randomly into normal control group, diabetic nephropathy group and treated group with CS (5.0 g/kg · d) for 2 weeks, 4 weeks and 6weeks. Then glomerular hypertyophy, renal function and 24h urinary, protein excretion were measured, and the expression of TGF-β and C-myc was examined by immunohistochemical technique. Finally we analyzed the relationship between TC, F-β and C-myc gene. Results :The level of serum ereatinine (SCr) , cholesterol (Chol) , proteinuria, serum TGF-β and collagen Ⅳ was all increased significantly in DN group, compared with that in control group ( P 〈 0. 05 ). It was found that glomerulus was corpulent, extracelluar matrix was increased, and renal interstitial was fiberized in mieroscopy. Meanwhile, after treating with CS STZ rats, the levels of proteinuria, SCr, Chol were reduced, and glomerular fibrosis was improved ( P 〈 0. 05). Otherwise, the overexpresssion of TGF-β and C-myc was significantly ameliorated by the treatment of CS for 6 weeks, compared with the DN group ( P 〈 0. 05 ). Conclusion: The gene expression of TGF-β and C-rove was significantly decreased after treating with CS. CS may contribute to the renoprotection in rats with DN.
