摘要
本文建立了快速分离与测定血清中甲硝唑及其代谢产物的反相高效液相色谱法。固定相为Shim-packCLC-ODS柱,以甲醇-水(27:73)作流动相,流速1.2ml/min,检测波长318nm。血样用甲醇除蛋白。甲硝唑平均回收率为99.6%,日内和日间相对标准偏差分别为3.7%和4.0%。最低检出浓度为0.025μg/ml,线性范围0.05~20μg/ml(γ=0.9999)。用本法研究了甲硝唑在大鼠体内的药动学。
reversed-phase HPLC method was developed for rapid separation and determination of metronidazole and itsmetabolite in rat serum using a Model LC-6A HPLC with two pumps and a Shim-pack CLC-ODC column. The mo-bile phase, mixture of methanol water (27: 73),was used at a flow rate of 1. 2ml/min. The serum samples repro-teinized with methanol and monitored at 318nm wavelength. The mean recovery of metronidazole in serum was99. 56%. Withinday and between-day variations were 3.7% and 4. 0%,respectively. The measurable lowest levelwas 0. 025μg/ml and the standard curve of metronidazole was linear over the concentration range of 0. 05 ̄20μg/ml (γ=0. 9999). The pharmacokinetics of metronidazole in rats after intragastric administration was studied.
