针刺对大鼠脑缺血超早期脑细胞内外游离钙离子浓度的影响

Effect of acupuncture on concentration of intra and extra cellular free calcium of the neuron in rats with(cerebral) ischemia in super early stage

目的:观察手十二井穴点刺放血法对大鼠脑缺血超早期脑细胞内游离C a2+浓度(〔C a2+〕i)和细胞外游离C a2+浓度(〔C a2+〕o)的干预作用,探讨其在中风急救中的作用机制。方法:30只W istar大鼠随机分为假手术组、模型组和治疗组。应用针型C a2+选择电极观测大鼠脑缺血即刻至20 m in期间每分钟脑缺血区皮质细胞〔C a2+〕o的变化。应用Fura 2/AM荧光探针技术观测脑缺血后20 m in脑缺血区皮质神经元突触体内胞浆〔C a2+〕i。结果:与假手术组比较,模型组大鼠造成脑缺血后,缺血区皮质细胞〔C a2+〕o从第5 m in开始出现明显降低(P<0.05),到20 m in为止其下降幅度与时间呈正相关(P<0.01),而缺血区皮质神经元突触体内胞浆〔C a2+〕i显著增高(P<0.01)。与模型组比较,治疗组大鼠脑缺血后缺血区皮质细胞〔C a2+〕o从第8 m in开始其下降幅度明显减小(P均<0.05),到第18 m in此趋势更加显著(P均<0.01),第20 m in缺血区皮质神经元突触体内胞浆〔C a2+〕i显著降低(P<0.01)。结论:脑缺血超早期应用手十二井穴点刺放血法进行干预可快速起效,起到调节缺血区脑细胞内外游离C a2+浓度,有效抑制神经元内钙超载,保护脑细胞功能的作用。

Objective： To observe the intervening effect of piercing and blood letting at twelve Jing points （十二井穴） of hands on the concentration of intra and extra cellular free calcium （[Ca^2＋]i and [Ca^2＋]o） of the neuron in rats with cerebral ischemia in super early stage, and investigate the mechanism of its neuroprotective effects on cerebral ischemic injury. Methods ： Thirty Wistar rats were randomly divided into sham-operated group, model group and treatment group. The [Ca^2＋]o in ischemia area per minute within 20 minutes after the ischemic injury was detected by the needle type Ca^2＋ selective pole. The [Ca^2＋]i of neuron in ischemic area at 20th minute after the ischemic injury was detected by using fluorescent indicator fura- 2/AM. Results.. Compared to the sham- operated rats, the [Ca^2＋]o of ischemic model rats decreased since 5 minutes （P〈0.05）, then the declination was positively correlated with time （P〈0.01） till 20 th minute of ischemia, while the [Ca^2＋]i increased obviously （P〈0.01）. Compared to the model group, the extent of [Ca^2＋]o declination in treatment group was restrained since 8 minutes （P〈0.05） and became more severe till the 18 th minute （P〈0.01）; at the 20 th minute, the [Ca^2＋]i decreased obviously （P〈0.01）. Conclusion： The piercing and blood letting at twelve Jing points of hands in treating cerebral ischemia in super early stage is able to rapidly regulate the [Ca^2＋]o and [Ca^2＋]i of neuron in ischemic area and restrains the over loading of calcium in the neuron. It has neuroprotective effects on cerebral ischemic injury.
