基因型和聚乙二醇预处理对甘蔗胚性愈伤组织原生质体分离的影响 被引量:1

摘要 对福农86/17、桂糖11号和粤糖57/423等3个甘蔗基因型的胚性愈伤组织进行了原生质体分离,研究了聚乙二醇(PEG)预处理对福农86/17胚性愈伤组织原生质体分离的影响.结果表明,桂糖11号的原生质体活力较高,但得率较低,福农86/17的原生质体活力和得率都较低,粤糖57/423的原生质体得率较高,但活力也较低.PEG预处理可以提高胚性愈伤组织分离的原生质体活力.一定质量浓度和一定时间的PEG预处理能够成倍地提高原生质体的得率,以质量浓度为0.50mg·L-1的PEG预处理15min。 Protoplasts were isolated from the embryogenic callus of 3 sugarcane genotypes, i.e. Funong 86/17, Guitang 11 and Yuetang 57/423. Effects of polyethylene glycol(PEG) pretreatment on protoplast isolation of embryogenic callus induced from Funong 86/17 was studied. The results showed that the protoplast vigor of Guitang 11 was comparatively high but the yield was comparatively low, both protoplast vigor and protoplast yield of Funong 86/17 were comparatively low, the protoplast yield of Yuetang 57/423 was high but the vigor was low. PEG pretreatment can raise protoplast vigor isolated from embryogenic callus. PEG pretreatment with given concentration and time can increase the protoplast yield several times as many. With the pretreatment of 50% PEG for 15 minutes, 1×10 6 live protoplasts were isolated and obtained from embryogenic callus per gram induced from Funong 86/17.
