颈淋巴结转移动物模型的建立及其微循环特性研究 被引量:2

摘要 为探讨血压变化对颈淋巴结转移瘤组织血流的影响,建立了颈淋巴结转移的动物模型,并测定了不同血压状态下转移瘤组织血流。给20只雄性Donryu大鼠耳廓背侧移植AH109A癌细胞14天后,使用氢气清除法测定不同平均动脉压状态下的组织血流量。结果20只大鼠中有17只发生同侧颈淋巴结转移。其肝、脑及骨髓的血流量当升压至20kPa(1kPa=7.5mmHg)左右或降压至9.06kPa左右时均无明显变化(P>0.1);而颈淋巴结转移灶的组织血流量在正常血压(14.81±1.04kPa)时为1.42±0.48ml·kg-1/s(n=13),升压状态(18.93±0.92kPa)下血流量增多至3.45±1.33ml·kg-1/s(n=13,P<0.05),降压状态(8.84±0.63kPa)下血流量骤然减少至0.55±0.25ml·kg-1/s(n=13,P<0.05),近乎于血流停止。结论:颈淋巴结转移瘤组织血流可随血压变化增多或减少。依此可望提高化疗效果。 Todeterminetheefectsofvasoactiveagentsonbloodflowinlymphnodemetas-tases,thebloodflowinthecervicallymphnodemetastasesandnormaltissueweremeassuredusingahydrogenclearancemethod,atnormotensivestate,angiotensinⅡ(AⅡ)inducedhypertensivestateandsodiumnitroprusside(SNP)inducedhypotensivestate.Themetastasesoccurredfrequentlyinthecervicallymphnodesof17ratsonday11~14afterYoshidaasciteshepatomaAH109Acelsweresubcutaneouslyinoculatedintobackofauriculeof20rats.Thebloodflowintheliver,brainandbonemarrowofnormalratswasofnosignificantchangeswhenthemeanarterialbloodpressure(MABP)waselevatedtoapproximately20kPabyinfusionofAⅡanddecreasedtoapproximately9.06kPabyinfusionofSNP.Butthebloodflowinthemetastaticlymphnodeswasincreasedfrom1.42±0.48ml·kg-1/sto3.45±1.33ml·kg-1/s(n=13,P<0.05)whentheMABPwaselevatedfrom14.81±1.04kPato18.93±0.92kPabyAⅡ.Onthecontrary,thebloodflowinthemetastaticlymphnodewasdecreasedto0.55±0.25ml·kg-1/s(n=13,P<0.05)whentheMABPwasde-creasedto8.84±0.63kPabySNP.Theresultsindicatedthatmicrocirculationcharacteristicsinthecervicallymphnodemetastaseswerediferentfromthoseinthenormaltissues.AⅡinducedhyper-tensionwasusefultoselectivelyimprovedeliveryofchemotherapeuticdrugstothesiteofmetas-tases.Ontheotherhand,SNPinducedhypotensionmaybeusefultosuppresswashoutofthedrugs.
