摘要
应用细胞培养技术,分子杂交的Northern Blot法和图像细胞分析技术(ICM),研究Ge—132对人乳癌细胞株BCaP—37的生物学作用。细胞生长曲线的结果显示,Ge—132的3个实验组(10.0mg/L,100.0mg/L,200.0mg/L)对细胞的生长没有显著抑制作用,并对细胞的毒性作用很小。基因检测显示,高浓度Ge—132(200.0mg/L)对癌基因C—erbB,和癌相关基因EGFR mRNA的表达具有抑制作用。ICM的四项指标显示,B和C两实验组(10.0mg/L,200.0mg/L)DI值呈异倍体。表现为恶性细胞的特征,两组核面积显著低于对照组(P<0.0l):C组的S期百分比显著低于对照组(P<0.05)。综合评价ICM各项指标发现,Ge—132(200.0mg/L)对细胞的增殖有一定抑制作用。
Biological effects of Ge-132 on Chinese breast cancer cell line--BCap-37 were studied by nieans of cell culture. Northern Blot and Iniage Cytonietry(ICM).Cell growth curves,showed that Ge-132(10.0,100.0,200.0mg/L)did not inhibit significantly the cell growth,and the toxicity of Ge-l32 to the cell was very low,The results of genc expresion indicated that Ge-132 in high concentration(200.0mg/L)inhibited C-erbB2 and EGFR mRNA expression, Four items were tested by ICM,DI in B and C groups showed the feature of malignant tumor cells. Nucleus areas in two groups(l0.0mg/L.,200. 0mg/L)were signifi-cantly lower than that in control group,and S%in C group(200.0mg/L)was significantly lower.The results of ICM suggestd that Ge-l32 inhibited significantly BCaP-37 cell proliferation by the general evaluation.
