BtMP—342菌株的生物学特性分析及毒力测定 被引量:1

摘要 1992年,从自然死亡尺蠖幼虫体内分离出BtMPk342菌株,经对落叶松毛虫室内外生测证明,该菌株是高毒力的优良菌株。1995年7月~8月期间,采用PCR技术分析了BtMPk342菌株特异性,该菌株含有CryⅠAa、CryAc、CryA、CryB以及Cry等五种毒蛋白基因。质粒电泳图谱分析表明,BtMPk342菌株由9个质粒构成,其中CryAa和Ac基因位于151.3Md和146.8Md以及48.7Md等质粒上。Cry和Cry均位于两个大质粒上。血清鉴定表明MP—342菌株属Kurstaki亚种,血清型为H3a3b。MP—342菌株在毒蛋白种类、质粒电泳图谱以及基因在质粒上的定位同HD—1菌株相比均有显著的不同。 BtMP-342 was separated from naturally died geometridae in 1992. The toxicity assay to Dendrolimus superans in laboratory and field indicated that the BtMP 342 has an excellently high toxicity. The specificity of BtM 342 was analyzed with PCR biological techniques from July to August in 1995. There were five kinds of toxic genes in BtMP 342, CryⅠAa, CryⅠAc, CryⅡA, CryⅡB, and CryⅤ. The electrophoresis analysis indicated that BtMP 342 was composed of 9 plasmids, of which, CryⅠAa, and CryⅠAc were located on 151.3 Md, 146.8 Md and as well as 48.7 Md, and both CryⅡ and CryⅤ were on two bigger plasmids. It was determined by serodiagnosis that MP 342 is a subspecies of kurstaki and its serotype was H3a3b. The differences in kinds of toxic protein, serodignosis and gene's location were obviously found between MP 342 and HD-1.
