固相合成肺表面活性物质蛋白质B的活性作用

摘要 为今后制备人工PS打下基础,用固相合成成熟段SP-B,用膜天平测定法和大鼠肺出血模型,观察它在体内、外的活性作用。结果:人工合成的磷脂加入合成SP-B在体外能降低表面张力至10mN/m以下,并增加滞后环面积;在体内能预防大鼠肺出血,使肺指数从0.73降至0.58(P<0.05),血清脂质过氧化物(nmol/L)从4.83降至2.74(P<0.01),肺泡血浆蛋白质(g/kg干肺)渗出从70.58降至21.98(P<0.01)。合用人工合成的SP-B优于单纯应用人工合成磷脂组,并与应用天然PS组相仿。 PURPOSE Pulmonary surfactant(PS) is an important material for maintaining lung function.Surfactant protein B(SP-B) is one of important components of PS.If the activity effect of synthetic mature SP-B is proved,it will be an foundation for preparation of synthetic PS which is very useful for clinical application METHODS The activity effects of solid-phase synthesized mature SP-B(S-SP-B) were observed with the mininum surface tension(γmin) and the area of hysteretic loop(Harea) in the membrane balance.Serum lipid peroxide(LPO),total protein(TP) in bronchoalveolar lavage liquid and lung index(LI) of pulmonary hemorrhage rats with or without S-SP-B were observed also.The analysis of variance was used.RESULTS When S-SP-B was added into synthetic phospholipids,the γmin was reduced to less than 10 mN/m and the Harea could be increased.The levels of TP(mg/g dry lung),LPO(nmol/L) and LI were decreased from 70.58,4,83,0.73 to 21.98(P<0.01),2.74(P<0.01),0.58(P<0.02) respectively.CONCLUSIONS The pulmonary hemorrhage could be prevented in rats with S-SP-B in combination with synthetic phospholipids and its effects was better than that in rats with synthetic phospholipids alone,or as similar as with natural PS.The effect of S-SP-B was proved in membrane balance also.
