Extraction and purification of the polysaccharides from Dunaliella salina(PDS) residue

摘要 目的从萃取过β-胡萝卜素后的盐藻藻渣中提取纯化盐藻多糖。方法采用热碱水提取,DEAE-纤维素和Sepharose-4B柱层析等方法分离纯化盐藻多糖。结果通过DEAE-纤维素柱层析分离得到3个组分,每个组分经过Sepdex-4B柱层析分别得到2个组分。结论这6个组分为均一组分,均为含有硫酸基和已糖醛酸的糖蛋白。 Objective To study the extraction and purification of polysaccharides from D. salina residue after extraction of β-carotene. Methods The crude polysaccharide was extracted by hot basic water, and then was isolated and purified by the DEAE-cellulose and Sepharose- 4B. Results After having been purified by DEAE-cellulose chromatography , three components named PDS1, PDS2, PDS3 were obtained. Each component was divided into two components by Sepharose-4B chromatography, then six components were obtained. Conclusion The six components were identified to be homogeneous polysaccharide-protein complexes which were composed of sulfate radical and uronic acid.
