角朊细胞免疫生物学研究

A STUDY OF IMMUNOBIOLOGY OF KERATINOCYTES

观察了人角朊细胞株Ｃｏｌｏ－１６细胞的免疫生物学特点。通过流式细胞术和细胞因子生物活性检测方法，研究了角朊细胞人白细胞抗原－ＤＲ（ＨＬＡ－ＤＲ）的表达和肿瘤坏死因子－α（ＴＮＦ－α）、白细胞介素－６（ＩＬ－６）的产生。结果表明，在重组人干扰素－γ（ｒｈＩＦＮ－γ）的作用下，角朊细胞ＨＬＡ－ＤＲ抗原的表达率为３１．８４％，２４ｈ角朊细胞培养上清中ＴＮＦ－α和ＩＬ－６生物活性分别为４６ｋＩＵ／Ｌ和２５ｋＩＵ／Ｌ，在ＬＰＳ，ＰＭＡ用ＩＬ－６刺激下，角朊细胞ＴＮＦ－α和ＩＬ－６产生显著增加。在细胞因子产生动力学研究中，也观察到在上述各种刺激作用下，角朊细胞ＴＮＦ－α和ＩＬ－６分泌峰值由７２ｈ提前到２４ｈ出现。提示角朊细胞具有重要的免疫学功能。

The immunobiological activity of Colo-16 cells, a human transformed keratinocyte cell line, were studied by using FACS (fluorescence-activated cell sorte) and WEHI-164 and 7 TD-1 bioassays. It was found that under the action of rhIFN γ(recombinant human interferon-γ), the rate of HLA-DR expression in cultured Colo-16 cells was 31.84％ in 3d,The bioactivities of TNF-α (tumor necrosis factor-α) and IL-6 (interleukin-6) in the supernatant of 24h Colo－16 cells culture were 46kIU／Land 25kIU/L respectively, and the productions and activities of them were significantly enhanced when the culture was stimulated by LPS (lipopolysaccharide) or PMA (phorbot myristate acetate) or IL－6. It was also found that in the kinetic study of the cytokines,the addition of the above stimulants would cause a left shift of the peaks of TMF-α and IL－6secretions from72h to 24h,These results suggest that the keratinocytes possess imporant immunological fuctions.
