摘要
观察了人角朊细胞株Colo-16细胞的免疫生物学特点。通过流式细胞术和细胞因子生物活性检测方法,研究了角朊细胞人白细胞抗原-DR(HLA-DR)的表达和肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的产生。结果表明,在重组人干扰素-γ(rhIFN-γ)的作用下,角朊细胞HLA-DR抗原的表达率为31.84%,24h角朊细胞培养上清中TNF-α和IL-6生物活性分别为46kIU/L和25kIU/L,在LPS,PMA用IL-6刺激下,角朊细胞TNF-α和IL-6产生显著增加。在细胞因子产生动力学研究中,也观察到在上述各种刺激作用下,角朊细胞TNF-α和IL-6分泌峰值由72h提前到24h出现。提示角朊细胞具有重要的免疫学功能。
The immunobiological activity of Colo-16 cells, a human transformed keratinocyte cell line, were studied by using FACS (fluorescence-activated cell sorte) and WEHI-164 and 7 TD-1 bioassays. It was found that under the action of rhIFN γ(recombinant human interferon-γ), the rate of HLA-DR expression in cultured Colo-16 cells was 31.84% in 3d,The bioactivities of TNF-α (tumor necrosis factor-α) and IL-6 (interleukin-6) in the supernatant of 24h Colo-16 cells culture were 46kIU/Land 25kIU/L respectively, and the productions and activities of them were significantly enhanced when the culture was stimulated by LPS (lipopolysaccharide) or PMA (phorbot myristate acetate) or IL-6. It was also found that in the kinetic study of the cytokines,the addition of the above stimulants would cause a left shift of the peaks of TMF-α and IL-6secretions from72h to 24h,These results suggest that the keratinocytes possess imporant immunological fuctions.
