双嘧达莫对细胞免疫的抑制作用 被引量:6

摘要 目的:观察双嘧达莫对小鼠免疫功能的影响。方法:采用小鼠末梢血白细胞计数,脾脏称重计算脾脏系数,测小鼠足垫厚度的差值确定小鼠迟发型超敏反应,用PHA和ConA诱生的脾淋巴细胞增殖测定淋巴细胞转化功能,作为活化的小鼠脾细胞测定小鼠IL-2的活性。结果:双嘧达莫1,10,40mg/kg抑制小鼠的迟发型超敏反应,降低小鼠末梢血白细胞计数和脾脏系数。双嘧达莫(Dip),1,10,100,1000μmol/L抑制PHA诱导的小鼠脾淋巴细胞转化和ConA诱导的小鼠脾淋巴细胞增殖反应,Dip1,10,100μmol/L亦抑制小鼠脾细胞产生IL-2。结论:实验结果为Dip老药新用提供一定的理论依据,临床上Dip应用于一些免疫性疾病的治疗似有较大潜力。 OBJECTIVE:To observe the effects of dipylidamole(Dip)on murine immune function. METHlD:Murine peripheral white blood cells count , and spleen index , measured by spleen weighing were usedto detectmurine foot pad thickness difference and to define the murine delayed hypersensitivity reaction. PHA and Con A in- duced murine spleen lymphocyte mitogenesis were used to determine lymphocyte transformation function. The acti- vating murine spleen cells were used to detect the activity of IL-2. RESULTS:Dip 1, 10, 40 mg/kg ip inhibitedmurine delayed hypersensitivity reaction induced by sheep red blood cells, and obviously decreased murine spleenindex and peripheral white blood cells number. Dip 1,10, 100, 1 000 μmol/L in vitro inhibited PHA induced murinespleen lymphocyte transformation and Con A induced murine spleen T lypphocyte cells mitogenesis. Dip 1,10, 00μmol/L inhibited interleukin-2 produced by murine splenocytes ( 1×106/ml). CONCLUSION:A theoretical basis isprovided for Dip, which will have potentialities in clinical application of some immunopathy therapy.
