摘要
目的研究肺癌患者血清中是否存在肺癌细胞分泌蛋白的特异性抗体。方法收集34例非小细胞肺癌患者血清、12例小细胞肺癌肺癌患者血清及15例健康对照者血清。收集A549细胞及HBE细胞培养的培养液上清液,同时收集胎牛血清培养液做对照;进行蛋白SDS-PAGE分离及电转膜。用肺癌患者及健康对照者的血清作第一抗体进行蛋白印迹检测。只与A549培养液蛋白样品有反应条带,而与HBE及对照无反应即认为是蛋白印迹阳性。结果在分子量约为30 kD的蛋白条带处,有61.8%(21/34)的非小细胞肺癌患者血清中存在分泌蛋白的抗体,小细胞肺癌患者(0/12)及健康对照者血清中未发现阳性条带(0/15)。结论非小细胞肺癌患者血清中存在分子量约为30 kD的A549分泌蛋白特异性抗体。
Objective To examine whether there was specific antibody to secreted protein of non-small cell lung cancer (NSCLC) cell line A549 in serum of patients with lung cancer. Methods The protein of conditioned medium of cell line A549, HBE and medium were recovered. The aliquot of proteins was subjected to SDS-PAGE, followed by Western blot analysis in which individual sera were tested for primary antibody. Sera from 34 newly diagnosed patients with NSCLC, 12 patients with small cell lung cancer(SCLC) and 15 healthy controls were analyzed. Results On band of molecular weight about 30 kD,sera from 61.8% of patients with NSCLC but none of the SCLC and healthy controls exhibited IgG-based reactivity against secreted proteins from A549. Conclusion There was specific antibody of secreted protein of A549 with molecular weight of about 30 kD in sera of NSCLC patients.
