铁超载与铁剥夺对HL-60细胞bcl-2和cmy-c基因的影响

摘要 目的探讨铁超载与铁剥夺对HL60细胞bc12和cmyc基因表达的影响。方法体外培养HL60细胞,分别加入不同浓度的三氯化铁(FeC13)。采用免疫组化LASB法测定凋亡相关基因bc12及cmyc表达。结果不同浓度的FeC13作用于HL60细胞6h、12h、24h、48h,bc12表达显著高于对照组。尤以100μmolLFeC13组显著,与对照组相比差异显著(P<0.05)。不同浓度DFO作用于HL60细胞一定时间后,其cmyc表达增高。结论铁超载时bc12高表达,铁剥夺时cmyc高表达。 Objective To investigate bcl-2 and c-myc Expression the Human Leukemic HL-60 Cells by iron, iron chelator. Methods HL-60 cells were cultured with different concentrations of iron ( Ⅲ )chloride (FeCl3) ,iron chelator-desferrioxamine(DFO) for 6h, 12h, 24h, 48h. They were divided into iron group (FeCl3 5, 10, 20, 50, 100,150μmol/L, 0 tmaol/L FeCl3 as the control group), DFO group(DFO 10, 20, 50, 100, 150μmol/L , 0 umol/L DFO as the control group). The expression of C-myc or Bcl-2 by immunohistochemistry methods. Results The results showed that when HL-60 cells were incubated with different concentrations of FeCl3 at 6h, 12h, 24h, 48h, the numbers of bcl-2 expression positive cells were much higher than that of control groups ( P 〈 0. 05). When HL-60 cell swere incubated with DFO, the expression of C-myc on HL-60 cells in DFO group was higher than that of control group at 6h, 12h, 24h( P 〈 0.05) .Conclusions Iron-overload could induced over-expression of bcl-2. Iron-deprived could induced upregulation of c-myc exoression in HL-60.
