z66抗体诱导伤寒沙门菌鞭毛抗原表达转变后染色体结构分析

摘要 目的 分析z66抗体诱导z66抗原阳性伤寒沙门菌鞭毛抗原表达改变后染色体结构。方法 利用脉冲凝胶电泳(PFGE)结合XbaI消化,对伤寒沙门菌标准菌株Ty2,z66抗原阳性株GIFU10007及z66抗体诱变j抗原阳性株GIFU10007-j的染色体DNA进行比较分析。结果 GIFU10007-j染色体DNA有两个XbaI片段与GIFU10007不同,总体染色体减少约250kb;与Ty2相比,GIFU10007染色体DNA有13个相同及7个不同的XbaI片段,总体染色体大小接近。结论 z66抗体诱导伤寒沙门菌鞭毛抗原表达转变后的染色体DNA发生了局部结构缺损;伤寒沙门菌z66鞭毛抗原阳性及诱变阴性株与单相标准菌株Ty2间在染色体DNA结构上有明显差异。 Objective To analyze the chromosome structure of Salmonella enterica serovar Typhi, which alternated flagelfin expression induced by the anti-z66 antiserum. Methods The pulse-field gel electrophoresis (PFGE) of the Xba I digested chromosome DNAs of S. enterica serovar Typhi Ty2, z66 positive swain GIFU10007 and GIFU10007-j induced by the anti-z66 antiserum from GIFU10007. Results Instead of a-270kb and a-30kb fragment of GIFU10007, a -45kb and a-28 kb fragment were appeared in the pattern of GIFU10007-j. There were thirteen identical fragments and seven different fragments between Ty2 and GIFU10007. Conclusion The flageUin expression of S. enterica serovar Typhi induced by z66 antibody is association with the partly deletion of chromosome DNA. The type strain Ty2 is different from the z66 antigen positive strain and the alternated negative strain in chromosome structure.
