摘要
目的:本研究旨在探讨干扰素-a(IFN-a)、白细胞介素-2(IL-2)、肿瘤坏死因子(TNF-a)单一、协同以及与化疗药物DDP的联合作用杀伤宫颈癌H e la细胞株的影响。方法:应用四M TT比色法检测与化疗药物DDP单独或联合应用组及药物作用的12h与24h的光密度值(OD),计算杀伤率。台盼蓝拒染计算杀伤率对照。结果:单独应用三种细胞因子时,IFN-a、TNF-a对宫颈癌H e la有细胞毒作用。IFN-a、IL-2、TNF-a联合作用具有协同作用,均与单一应用具有显著性差异(P<0.01)。细胞因子均可以增强顺铂(DDP)的抑瘤作用并与其作用的先后及时间具有依赖关系。结论:M TT比色法为临床肿瘤化疗的药物敏感性检测提供了简便、快速的方法。不同类型的肿瘤有着不同的药敏谱。干扰素-a(IFN-a)、肿瘤坏死因子(TNF-a)、白细胞介素-2(IL-2)与DDP联合具有协同作用,为宫颈癌病人化疗提供了理想的参考方案。
Objective: To investigate the effects of rhIFN-a, rhIL-2 and rhTNF-a on the Hela cell line in vitro in a single or cooperative form or in combination with chemotherapeutics DDP. Methods: Chemosensitivity of the cells to doxorubicin was analysed using the MTT assay 12h and 24h after using rhIFN-a, rhIL-2 and rhTNF-a alone and in combination with chemotherapeutics DDP,and compared by using typanlan blue method. Results: When the three factors were used alone, IFN-a or TNF-a there had toxic action on the uterine cervix cancer Hela cell line , and when they were used cooperatively, there was significant difference in the effects on the Hela cell line( P〈 0.01 ). All the cytokines had the enhancement of DDP antitumor activity in vitro with the dependence of the antitumor effect on their action order and time. Conclusion: The MTT assay is very rapid and simple. Chemosensitivity spectra vary with tumor types, rhIFN-a, rhIL-2 and rhTNF-a have the synergistic inhibition in combination with DDP, which provides provide better chemotherapy choice for patients with uterine cervix cancer.
