摘要
目的测定唐古特大黄多糖中单糖的组成及其物质的量之比。方法将大黄多糖样品用2 mol.L-1的硫酸溶液降解成单糖后,用1-苯基-3-甲基-5-吡唑啉酮衍生化成单糖,应用毛细管区带电泳法测定各单糖。熔融石英毛细管柱(50μm×70 cm,有效长度60 cm);二极管阵列检测器;运行缓冲液为硼砂水溶液(浓度为150 mmol.L-1、pH值为10.8);分离电压10 kV;柱温25℃;0.5 Pa压力下进样,进样时间5 s。结果唐古特大黄多糖由木糖、阿拉伯糖、葡萄糖、鼠李糖、半乳糖、甘露糖、葡糖醛酸、半乳糖醛酸等8种单糖组成,其物质的量之比为1.00∶23.27∶15.93∶2.95∶1.34∶49.07∶2.62∶4.31。结论该方法具有简单、快速、分离效率高等特点,可用于唐古特大黄多糖中单糖组成的测定。
Objective To determine the monosaeeharides in rheum tangutieum maxim polysaeeharides and their molar ratio. Method The rheum tangutieum maxim polysaeeharides were hydrolyzed into monosaeeharides with 2 mol·L^-1 sulfuric acid. The monosaeeharide derives obtained with 1-phenyl-3-methyl-5-pyrazolone (PMP) were determined by capillary zone eleetrephoresis (CZE) with fused silica capillary tube (70 cm × 50 μm, effective length is 60 era), diode-array detector, 150 mmol·L^-1 sodium tetraborate buffer ( pH value was 10.8 ), 10 kV, 25 ℃, and 0.5 psi pressure injection for 5 s. Results Eight monosaeeharides derivatives were isolated with high sensitivity, good resolution by CZE. The rheum tangutieum maxim polysaeeharides are composed of xylose, arabonose, glucose, rhamnose, galaetose, mannose, glueuronie acid and galaeturonie aeid with a molar ratio of 1. 00 : 23.27 : 15.93 : 2.95 : 1.34 : 49.07:2.62 : 4.31. Conclusion This method is simple, rapid, sensitive and can be used to determine the monosaeeharides in rheum tangutieum maxim polysaeeharides.
