具有高再生性且对农杆菌C58敏感的玉米自交系材料的筛选 被引量:1

摘要 以70个优良高代玉米自交系的幼胚为材料进行离体培养,从中筛选出胚性愈伤组织诱导率高、克隆力强的10个自交系,将其一部分胚性愈伤组织用作分化,研究其再生能力;另一部分用农杆菌C58转化,利用植物载体中携带的G US报告基因的瞬时表达研究玉米基因型对农杆菌的敏感性。结果表明,10个自交系都具有再生能力,自交系6010、6060、6038、6051和6015的分化力相对较高。玉米基因型对农杆菌的敏感性存在极显著差异,其中自交系6051最敏感,而6034、6038对农杆菌不敏感。因此认为自交系6051、6015、6010和6060是再生能力强且对农杆菌C58敏感的自交系材料。 10 maize inbred lines with high induction efficiency of embryonic callus and embryonic callus clone ability have been selected from 70 maize inbred lines by immature embryo culturing. To study generation of embryonic callus initiated from 10 maize inbred lines some embryonic callus were transferred to shoot-developing media to generate. The others were transformed via Agrobacterium tumefactions C58 containing GUS reporter gene to study the susceptibility of different genotypes in maize to Agrobacterium tumefactions using GUS gene transient expression. The results showed that all the embryonic callus initiated from 10 maize inbred lines were able to generate, and the generation of inbred lines 6010,6060,6038,6051 和 6015 are higher compared with that of the others. There significantly differed in the susceptibility of different genotypes in maize to Agrobacterium tumefactions, and all genotypes were susceptible to Agrobacterium tumefactions C58 but inbred lines 6034 and 6038. So inbred lines 6051,6015,6010 和 6060 were thought as materials with high generation and the susceptibility to Agrobacterium tumefactions C58.
