整合素α_5及整合素β_3在子宫内膜异位症中表达的研究 被引量:1

摘要 目的:探讨整合素α5、β3在子宫内膜异位症(EM)患者在位和异位内膜中的表达。方法:采用逆转录聚合酶链反应(RT-PCR)技术,对18例EM患者在位内膜、异位内膜(研究组)和18例非EM患者子宫内膜(对照组)行整合素α5、β3mRNA检测。同期放射免疫法测定EM患者体内雌二醇(E2)、孕酮(P)、糖类抗原125(CA125)水平,对EM患者术中行美国生殖学会评分系统(AFS-r)评分。结果:整合素α5mRNA在EM患者在位内膜、异位内膜及对照组子宫内膜中表达水平分别为0.773±0.113、1.077±0.032、0.924±0.120,各组间比较差异具有统计学意义(P<0.05);整合素β3mRNA在EM患者在位内膜、异位内膜及对照组子宫内膜中表达水平分别为0.653±0.080、0.963±0.132和0.763±0.090,各组间比较差异具有统计学意义(P<0.05)。EM组血清E2水平与异位内膜整合素α5、β3mRNA表达均呈正相关(P<0.05),相关系数分别为0.793、0.917;而P水平与异位内膜整合素α5、β3mRNA表达均无相关(P>0.05)。CA125水平与异位内膜整合素α5、β3mRNA表达均呈正相关(P<0.05),相关系数分别为0.763、0.754,AFS-r评分与异位内膜整合素α5、β3mRNA表达均呈正相关(P<0.05),相关系数分别为0.801、0.773。结论:EM异位内膜组织中整合素α5、β3高表达可能与EM黏附发生有关,整合素α5、β3表达受雌激素调控。 Objective : To study the expression of integrin α5, β3 in eutopic endometrium and ectopic endometrium of endometriosis. Methods: Reverse transcription polymerase chain reaction (RT-PCR) method was utilized to detect and quantitate integrin α5, β3 mRNA expression levels in eutopic and ectopic endometrial tissues from 18 patients with endometriosis and the endometrial tissues from 18 non-endometriosis women. Serum concentration of estradiol ( E2 ) , progesterone (P) and CA125 were measured by radioimmunoassay. AFS-r marking was used to describe the state of pelvic cavity during operation. Results : Various levels of positive expression of integrin subunits α5, β3 were shown in both eutopic and ectopic endometrial tissues of endometriosis and the endometrium of non-endometriosis. The expressions of integrin α5 mRNA in um of control group we groups, the differences eutopic endometrium, ectopic endometrium of endometriosis group and endometrire as 0.773 ±0. 113, 1.077 ±0.032, 0.924 ±0. 120, respectively. Among the were statistically significant ( P 〈 0.05 ). The expression of integrin β3 mRNA in eutopic endometrium, ectopic endometrium of endometriosis group and endometrium of control group were as 0.653 ± 0. 080, 0. 963 ± 0. 132, 0. 763 ± 0. 090, respectively. Among the groups, the diferences were statistically significant ( P 〈 0.05 ). Serum concentration of E2 in study group was positively correlated with integrin α5, β3 (r=0. 793, 0. 917 ,P 〈0.05). There were no relations between serum concentration of P and integrin α5, β3 (P 〉0.05). Both CA125 level and AFS-r were positively correlated with integrin α5, β3 ( P 〈 0. 05 ). Conclusion : The high expression of integrin α5, β3 in ectopic endometrium might play an important role in the development of endometriosis adherence . Estradiol may regulate the expression of integrin α5, β3.
