摘要
用MTT法研究不同浓度的氯化镉对中国地鼠肺细胞(CHL)的细胞毒性,结果表明:MTT还原量减少,甲生成量与氯化镉浓度、染毒时间之间存在依赖关系,24和48小时的半数生长抑制浓度(IC50)分别为5.7×10-6和1.66×10-6mol/L。我们还研究了不同浓度的氯化镉对CHL细胞的染色体畸变效应,结果表明:不同浓度氯化镉引起的CHL细胞染色体畸变率显著高于阴性对照组,畸变类型主要包括染色体断裂、易位、多倍体等。畸变率的增加随氯化镉浓度增加和染毒时间的延长而逐渐升高。
The cytotoxicity of CdCl2 with CHL cells by MTT method was studied. The results showed that the reduction of MTT was decreased,and the amount of formazane produced depended on the concentration of CdCl2 and the time of intoxication. The 50%inhibition (Ic50) of cadmium chloride for 24 and 48 hrs. was achieved at 5.7μM and 1.66μM respectively. At the same time,the effects of CdCl2 on chromosome aberration were also studied.The results showed that the frequency of chromosome aberration in the treatment group was obviously higher than that of the control group,and the type of aberration included break, translocation and polyploid, etc. The increase of aberration frequency was gradually elevated as the CdCl2 concentration increased and the time of intoxication elongated.
