CpG ODN对rHBsAg免疫小鼠NK细胞表面分子表达及其活性的影响

摘要 目的探讨合成的含CpG基序的寡核苷酸(CpG ODN)与重组乙型肝炎病毒表面抗原(rHBsAg)联合免疫小鼠对NK细胞表面标志NK1.1表达情况及其细胞毒活性的影响。方法采用C57BL/6小鼠,经后腿胫骨前肌免疫2次,FACS检测脾细胞NK1.1表面分子的表达,生物活性法测定脾脏NK细胞的细胞毒活性。结果各实验组间NK细胞表面标志NK1.1的表达差异无显著意义;加CpG ODN各组与相应未加CpG ODN组比较对靶细胞的杀伤活性显著增强,且各组与对照组比较杀伤活性差异均具有显著意义。结论CpG ODN对免疫小鼠NK细胞表面分子NK1.1的表达无明显影响,而对其细胞毒活性具有明显的增强作用,显示CpG ODN具有潜在的免疫佐剂效应。 Objective To explore the influence of synthetic oligodeoxynucleofide containing CpG motif(CpG ODN) on the surface marker NK1.1 expression and cytotoxicity of NK cells of mice immunized with rHBsAg. Methods Divide C57 BI/6 mice into five (i. e. control, rHBsAg, rHBsAg + Clog ODN, vaccine and vaccine + Clog ODN) groups, five for each. Each mouse was injected with Clog ODN and/or the corresponding antigen in back leg fibialis anterior(TA) for 2 times, at an interval of 4 weeks. Determine the expression of NK1.1 by FACS and the cytotoxicity of NK cells by bioactivity assay. Results No significant difference was observed in the expression levels of NK1.1 in the four test groups. However,compared with those of mice unimmunized with Clog ODN, the cytotoxicity of NK cells of mice immunized with Clog ODN increased significantly,and the cytotoxicity in the four test groups were significantly higher than that in control group. Conclusion Clog ODN showed no significant influenee on the expression of surface marker NK1.1 of NK cells of mice immunized with rHBsAg, however, it significantly enhanced the cytotoxicity of NK e.ells of the mice. It indicated the potential adjuvant effect of Clog ODN.
