细粒棘球绦虫细胞体外培养的研究Ⅰ.细粒棘球蚴生发层细胞分离

摘要 本文报道采用胰酶消化的方法分离细粒棘球蚴生发膜细胞的研究结果,在37℃下10分钟的效果最好,延长消化时间可使细胞浆破裂,形成较多的裸核,不利于细胞进一步培养。梯度离心的分离方法增加细胞损伤,不能达到分离不同细胞的效果。分离的生发层细胞超微结构与完整生发膜超微切片上基本相同,能表现有线粒体肿胀和数量减少等特点,表明胰酶处理的细胞代谢能力受到一定的抑制。将分离的生发层细胞接种Balb/c小鼠腹腔可形成棘球蚴,并产生成熟的原头节。讨论了分离方法对生发细胞体外培养工作的意义。 This paper reports the result of isolation and charaterization of the germinal cells from hydatidcysts with the method of trypsin digestion. The best result was obtained at 37 ℃ for 10 minutes. The cytoplas-ma were broken and more naked nuclei were formed when the time for digestion was prolongated. Separationby gradient centrifugation increased cell trauma and could not separate the different cells. The fine structureof the cells isolated were identical to that of the cells of intact germinal niembrane on ultrathin section. Butthe swollen and hydropic mitochondria were found and the number of mitocliondria decreased in isolated cells.It suggested that the metabolism and energy supply of trypsin-treated cells were inhibited to a certain extent.The metacestode and mature protoscoleces developed after inoculation of isolated germinal cells into Balb /Cmice. The significance of methods used in cell isolation is discussed.
