骨髓增生异常综合征患者骨髓DC1亚群的变化 被引量:4

摘要 目的:探讨树突状细胞1型(DC1)存骨髓增生异常综合征(MDS)发病中的作用。方法:以正常人和重型再生障碍性贫轿.(SAA)为埘照,用流式细胞仪检测 MDS、SAA 骨髓中 Th1细胞数、DC1亚群-CD11_+CD1~a^+细胞、CD11_^+CD83^+细胞数量及比例改变;评价 CD11_+CD83^+细胞与 Th1细胞、网织红细胞(Ret)、中性粒细胞绝对值(ANC)的相关性。结果:与正常人比较,MDS 患者 Th1细胞及 DC1亚群均未见增高(P<0.05),CD11_+CD83^+细胞与 Th1细胞、ANC 无显著相关性(P>0.05).但与 Ret 呈正相关(P<0.05)。SAA 的 Th1细胞、CD11_+CD1_a^+细胞、CD11_+CD83^+细胞、CD11_+CD83^+细胞/CD11_+CD1_a^+细胞均显著高于正常对照组(P<0.01)。SAA 患者 CD11_+CD83^+细胞数与 Th1细胞呈正相关(P<0.01),与 Ret、ANC 均呈负相关(P<0.01)MDS 的 CD11_+CD1_a^+细胞、CD11_+CD83^+细胞、CD11_+CD83^+细胞/CD1_a^+CD11_+细胞均低于 SAA(P<0.01)。结论:MDS 患者 DC1亚群处于相对静息状态.但激活 DC1可能有助于 MDS 造血恢复。MDS 和 SAA 的免疫发病机制可能不同。 Objective To evaluate the changes of dendritic cells 1 (DC1) subsets in the bone marrow of patients with myelodysplastic syndrome (MDS). Methods The quantities of the CD11c^+CDla^+ cells, the CD11c^+CD83^+ cells and the Thl cells in the bone marrow of MDS patients, severe aplastic anemia (SAA) patients and normal controls were detected respectively by flow cytometry. The relationships between the CD11c^+CD83^+ cell counts and the Thl cell counts, the reticulocytes (Ret) and the absolute neutrophil count (ANC) were evaluated respectively. Results There was no significant difference in the Thl cell counts, the CD11c^+CD1a^+ cell counts, the CD11c^+CD83^+ cell counts and the ratio of CD11c^+CD83^+/ CD11c^+CD1a^+ between normal controls and MDS patients. The CD11c^+CD83^+ cell counts were positively correlated with Ret in MDS (P〈0.05). The CD11c^+CD1a^+ cell counts, the CD11c+CD83^+ cell counts and the ratio of CD11c^+CD83^+/CD11c^+CD1a^+ in MDS patients were lower than those in SAA patients (P〈0.01). The CD11c^+CD83CD11c^+ cell counts were positively correlated with Thl cell counts (P〈0.01), and negatively with Ret and ANC (P〈0.01) in SAA. Conclusions The immunity of DC1 in MDS was in a relatively quiescent state. The activated DC1 might facilitate the recovery of the hematopoiesis in MDS. The immunological pathogenesis of MDS might be different from that of SAA.
