痉证及痿证中药方对大鼠脊髓持续性压迫损伤局部ET-3mRNA表达的影响 被引量:2

摘要 目的:研究大鼠脊髓持续性压迫后缩血管因素ET-3mRNA的改变,探讨痉、痿证方对大鼠脊髓持续性慢性损伤局部ET-3mRNA表达的影响。方法:将雄性中年大鼠随机分为假手术组、轻压、中压及重压模型组和轻、中、重压的痉证方、痿证方治疗组。采用大鼠颈椎前路螺钉压迫颈脊髓,螺钉留置持续压迫30d以产生慢性损伤,用RT-PCR方法检测受压局部ET-3mRNA表达变化。结果:正常脊髓组织中有ET-3mRNA表达,脊髓受压后ET-3mRNA表达明显增高,模型组与假手术组相比各组均P<0·01;ET-3mRNA表达增高程度与压迫程度有关,与轻压组相比,中、重压组差异具有显著性意义,P<0·01,重压与中压组相比,差异具有显著性意义,P<0·01;痉、痿证方干预后,增高的ET-3mRNA明显降低,与模型组相比,痉、痿证方组有统计学差异,P<0·01。结论:痉、痿证中药方对增高的ET-3mRNA有明显的降低作用。 Objective: To study the alteration of ET-3 mRNA expression in rats' spinal cord after constant compressive injury, furthermore to explore the effects of J ing syndrome recipe (痉证方)and Wei syndrome recipe(痿证方)on ET-3 mRNA expression in injury local of spinal cord in rats. Methods:The midlife male rats were randomly divided into four groups:sham-operated group, model groups which included mild, moderate and severe compress groups, therapy groups of Jing syndrome recipe(痉证方) and Wei syndrome recipe(痿证方) which also included mild, moderate and severe compress groups. Screws were used to drill into spinal cord through anterior cervical vertebra body. After operation the screws remained in spinal cord for 30 days to create chronic injury, and then RT-PCR was used to check the expression changes of ET-3 mRNA in compressed location. Resets:There was expression of ET-3mRNA in normal spinal cord tissues. After spinal cord was com- pressed, the expression of ET-3mRNA increased significantly( P 〈 0.01 ). The increase degrees of ET-3mRNA expressions were related to the degree of compression, and the expressions were different in the mild, moderate and severe compress groups( P 〈 0.01 ). After Jing syndrome recipe (痉证方) and Wei syndrome recipe (痿证方) were used, the increased ET-3mRNA expression decreased significantly( P 〈 0.01 ). Condmion: Jing syndrome recipe(痉证方) and Wei syndrome recipe(痿证方)are effective to decrease ET-3mRNA expression in injuried spinal cord after constant compression.
