摘要
以新透心红胡萝卜为植物材料,用微弹轰击法对胡萝卜愈伤组织进行了降钙素基因的遗传转化。对所得再生植株进行了PCR检测及Southern印迹检测。在16棵Kan抗性株中有13棵PCR阳性株;4棵存活的PCR阳性株经Southern印迹检测,有3株为阳性。对收获的2个肉质根做PCR检测,均为阳性;对其中较大的肉质根作Southern印迹检测,结果也为阳性。这些检测结果说明降钙素基因被成功地整合到胡萝卜基因组中。
Carrot cultivar 'Xintouxinhong' was transformed with ealeitonin gene by micro-bombardment. Among 16 Kanamycin-resistant plants, 13 were positive by PCR detection. 3 plants were positive by Southern blot detection in 4 living plants. Two fleshy roots were harvested in the 3 positive plants and they showed positive in PCR detection. Southern blotting of the bigger one fleshy root showed it was positive. The results indicated that the calcitonin gene had been inserted into the carrot genome DNA.
