摘要
乙酰胆碱酯酶是胆碱能神经传导中的一种关键酶,对其构效关系的研究一直是生命科学的重要课题,目前主要研究手段有X线衍射晶体学、分子生物学、电子顺磁共振等。本文重点介绍应用这些方法研究乙酰胆碱酯酶构效关系所取得的新进展,包括芳香族引导、后门开放、环的纳秒机制与高效催化的关系等。
Acetylcholinesterase (ACHE) is one of the key enzymes in cholinergic nervous system. The relations between its structure and function continues to be the important subject of the study in many fields of life sciences. At present, the methods to study the structure-function relations of AChE mainly include X-ray crystallography, molecular biology, electron paramagnetic resonance and so on. In this brief review, the advances obtained by these technologies in the research on the structure-function relations of AChE were introduce, including the relations of the high catalytic efficiency with the aromatic steering, back-door mechanism, the Ω-loop regulation and so on.
